INSILICO to Present First-in-Human Phase I Data for AI-Designed Pan-TEAD Inhibitor ISM6331 at ESMO 2026

InSilico Medicine Cayman TopCo (INSILICO) has announced that Phase I clinical data for its novel pan-TEAD inhibitor, ISM6331, have been selected for a Rapid Oral presentation at the European Society for Medical Oncology (ESMO) Congress 2026, scheduled for 25 October 2026 in Madrid, Spain.

The multicenter, first-in-human study (Presentation No. 1004RO) is evaluating ISM6331 as a single agent in patients with advanced or metastatic malignant mesothelioma and other solid tumors. Conducted concurrently in China and the United States, the trial assesses safety, tolerability, pharmacokinetics, pharmacodynamics and preliminary anti-tumor activity.

ISM6331 is an AI-designed, non-covalent pan-TEAD inhibitor created using Insilico’s Chemistry42 structure-based drug design platform. Preclinical studies showed broad anti-tumor efficacy at low doses and a favorable safety and ADMET profile. The U.S. Food and Drug Administration granted ISM6331 orphan drug designation for mesothelioma in August 2024, and first patient dosing in the global Phase I trial began in January 2025.

The Board of INSILICO highlights that the upcoming ESMO presentation represents a significant validation of the program’s clinical progress.
